DNA is a string over A, C, G and T. The complement pairs A with T and C with G; a reverse complement also reverses orientation. A substitution, insertion or deletion can change sequence features without proving a phenotypic effect. Verify sequence direction and reference coordinates before comparing variants.
Worked example
For 5'-ATGC-3', the complement is TACG and the reverse complement is GCAT. A single base change can be counted before asking whether it alters a coding region.
